Depends on the number of generations you want to get w/ genetic defects. IIRC, you only need about 30 women and only a handful of men to maintain the population, but that certainly will cause problems in later generations.
I don't know where I got it from, but a population of min 100,000 individuals would be viable. Now if the man wants to lead a "normal" life instead of breeding 24/7, I've read somewhere that he could impregnate about 4,000 women - do the math and start the elimination process
